Gene Runner needs to be installed prior to its launch. Download the installation package, then you'll find the shortcut on your desktop. Double click it and the main interface will open, then depending on your needs, go to File > New and choose a sequence to start it.

Gene Runner is not available for Mac. However, there are other programs you can use for Mac, such as ApE or SnapGene.
